One note: it won’t even let you update over 3G (the button is grayed out) – WiFi Only!

“To get this update, you’ve only to plug your device in to your computer and start up iTunes, hit your device on the side bar, and find the Version row under Summary.” If you have iOS 5 already, you just need to plug your phone into a charger and go to Software Update in the General Settings. No PC needed.

If you already have iOS 5 installed, then you don’t need iTunes to update. Just go to Settings->General->Software Update to update, untethered. The iOS 5.0.1 update appears to be ~55 MB, so best to do this over Wi-Fi.
